Papanicolaou Stain EA-65 is an alcohol-based cytoplasmic counterstain formulated for use in the Pap staining procedure. This polychromatic solution contains Eosin Y, Light Green SF Yellowish, and Bismarck Brown to provide strong, balanced cytoplasmic differentiation. EA-65 is designed to produce more intense staining than EA-50, making it well suited for thicker or more keratinized cytology specimens. When used after hematoxylin nuclear staining and bluing, it functions as a counterstain to clearly distinguish cellular components. Non-keratinized squamous cells stain blue-green, while keratinized or mature cells appear pink to orange, improving contrast and aiding microscopic interpretation. Ideal for clinical, hospital, and reference laboratory cytology applications.
